Lentiviral vectors are commonly used in the production of ex vivo cell therapies to transduce various cell types. Lentiviral vectors are sensitive to the shear forces of traditional continuous centrifugation systems. Additionally, significant yield losses due to adsorption effects can be seen during the primary clarification of lentiviral vectors with single-use filtration methods such as depth filtration.

Depth filtration has become a trusted standby for scientists and engineers looking to separate viral vectors – a popular delivery system for gene-based therapies and vaccines – from the HEK293 cell culture in which they’re typically grown. In this case study, we show how the UFMini single-use centrifuge significantly reduces filtration needs and drastically enhances the amount of viral titer collected when compared to conventional methods.
